Here is what I did for my 55 gallon tank:

Tank
Rapids Wholesale - Co2 kits - 5-lb new steel tank with dual gauge regulators
$99 (#BD435)

Needle Valve
Florida driftwood - needle valve used in medical gases $19.50
Appears to be Clippard MNV-4K2 (4k2 has an 1/8 hose barb included)

Also needs a ¼” NPT male to 1/8” NPT female adaptor (Clippard #15006-5) to connect the valve directly to the regulator.
Can probably get where you fill co2 gas.

Note some online aquarium stores now carry the larger version of this, which is what I have.

NOTES

You may be able to buy the clippard needle valve - MNV-4K2 from a local Clippard distributor. Check their web site

Some people use other co2 reactors/diffusers. Lots of variety here so do some more research on the web.

My 5 lb tank lasts about 8 months.

Or you could get the needle valve from Aqua Botanic and it comes with the adaptor for $18.

And - in addition to what Rex said -
1) If you find a local Clippard distributor you can likely get the valve for $11; and maybe whatever adaptors you need for free.

2) If you shop around at local welding supply places you might beat the price on the tank and regulator.

Be sure to line up your source of CO2. Some places will only exchange tanks. If so, no need to buy a brand new one and especially not an aluminum one. I get my CO2 from a fire extinguisher supply company.

Try looking at a homebrew store for your tank and regulator. They may be able to fill your tank also.

Those Dupla CO2 reactors work by the level of CO2 reacting with the air inside the reactor correct?
There is a liquid you add which changes in ph as the tank does correct?

I have a similar one from Aquarium Products.

I added the Dupla CO2 monitor after I changed my filter and tryed to eyeball/tweak the co2 gas flow without counting bubbles. Bad idea. At least now everytime I walk by the tank, I can easily glance at the monitor. Green = good.
