Does your stand have a long flat base, or does it have feet like a table? Assuming the long flat base, I would set up the tank to run counter to the supports in the floor. On 16" centers, a 2' deep tank will be supported by no more than 2 beams, and probably only 1. If you turned it across the beams, then the load will spread across 3 beams wherever you put it. It also helps to put the tank as close as possible to over a load bearing wall on the first floor. The farther you get from the support, the more the beam will deflect.

If your stand has feet, you might consider making a load spreading frame that the feet will sit into and transfer the load from the points under the feet across the beams in the floor.
