Default How much mulm?

So how much mulm do you add to the substrate when you set up a new tank? Do you mix it or layer it into the botton under the substrate? Does it matter?

My new 50 goes up this wkd and being the largest tank I have ever set up, want to minimize early mistakes.

As much mulm as you can scrape up, more the better, place it in first, then sub over, I have done several tanks this way, and each one have done flawlessly, never a bloom of any kind.
